摘要

目的：建立测定维U颠茄铝胶囊Ⅱ中硫酸天仙子胺和东莨菪内酯含量的方法，并考察相关影响因素的影响。方法：采用高效液相色谱法检测，用十八烷基硅烷键合硅胶为填充剂，以0.1 mol·L-1庚烷磺酸钠溶液为流动相A：甲醇为流动相B，梯度洗脱，流速为1.0 mL·min-1，柱温为35 ℃，检测波长为210 nm（硫酸天仙子胺）和344 nm（东莨菪内酯），进样量为100 μL。结果：硫酸天仙子胺和东莨菪内酯的线性范围分别为4.098～40.980 μg·mL-1和0.209 7~2.097 0 μg·mL-1，回收率为分别为100.3%和100.7%（RSD为1.9%和2.8%，n=6）。硫酸天仙子胺和东莨菪内酯在热环境下不稳定，且随着温度的升高，含量下降越明显。结论：建立的方法可准确测定维U颠茄铝胶囊Ⅱ中硫酸天仙子胺和东莨菪内酯的含量，专属性好，前处理简单。影响硫酸天仙子胺和东莨菪内酯含量的主要因素为温度和时间，提示生产企业应注意控制生产过程中干燥工艺的温度和时间，同时应控制好贮存条件，避免在流通环节温度过高引起产品质量不合格。

Abstract

Objective： To establish a method for determining the content of hyoscyamine sulfate and scopolamine lactone in Vitamin U Belladonna Aluminum Capsules Ⅱ， and to investigate the influence of influencing factors. Methods： High performance liquid chromatography was used for detection， with octadecylsilane bonded silica gel as the filler， 0.1 mol·L-1 sodium heptanesulfonate solution as the mobile phase A， methanol as the mobile phase B， gradient elution， flow rate of 1.0 mL·min-1， column temperature of 35 ℃， detection wavelengthes were set at 210 nm (asparagine sulfate) and 344 nm (Scopolamine lactone)， and injection volume of 100 μL. Results： The linear ranges of hyoscyamine sulfate and scopolamine lactone were 4.098-40.980 μg·mL-1 and 0.209 7-2.097 0 μg·mL-1， respectively， and the recovery rates were 100.3% and 100.7%， respectively (RSD of 1.9% and 2.8%， n=6). Hyoscyamine sulfate and Scopolamine lactone were unstable in hot environments， and their contents decreased more significantly with increasing temperature. Conclusion： The established method can accurately determine the content of hyoscyamine sulfate and scopolamine lactone in vitamin U belladonna aluminum capsules Ⅱ， with good specificity and simple pretreatment. The main factors affecting the content of sulfamethoxazole and scopolamine lactone were temperature and time. It was suggested that production enterprises should pay attention to controll the temperature and time of the drying process during the production process， and at the same time， control the storage conditions to avoid excessive temperature in the circulation process， which may cause product quality non-compliance.
