Abstract: Objective: To clarify the material basis of the yin-nourishing effect of rehmannia radix and the correlation between efficacy and mechanism, to establish a multi-component qualitative identification method under the same thin-layer chromatography (TLC) conditions, and to confirm the transferability of its characteristic components in the compound preparation. Methods: Based on network pharmacology studies, the active components with the yin-nourishing effect in rehmannia radix were predicted. TLC method for simultaneous qualitative identification of multiple components of rehmannia radix was established, with the chromatographic conditions being: high-performance silica gel G thin-layer plate, 12∶8∶1 of chloroform-methanol-water as the developing solvent, and 5% vanillin-sulfuric acid solution as the chromogenic agent. The transfer of target components in the compound preparation was traced under the same chromatographic conditions. Results: Based on network pharmacology, catalpol, rehmannioside A, rehmannioside D, and ajugol were confirmed as the characteristic components with the yin-nourishing effect in rehmannia radix. A method for qualitative identification of 4 characteristic components of raw rehmannia radix and 2 characteristic components of prepared rehmannia radix was established, and the transferability of the characteristic components in the compound preparation was determined. Conclusion: The quality markers screened out by network pharmacology can be used to clarify the material basis of the efficacy of rehmannia radix, and the thin-layer identification method established realizes multi-side (4 components) qualitative identification of characteristic components of rehmannia radix, which provide methodological basis for the quality evaluation of various chemical components of rehmannia radix medicinal materials and rehmannia radix compound preparations.
