摘要: 目的:建立以羧甲基-β-环糊精(CM-β-CD)为手性流动相添加剂的高效液相色谱法(HPLC),对美托洛尔、文拉法辛两种手性药物进行对映体拆分研究。方法:色谱柱为SunFire TM C18柱(4.6 mm?15 mm,5 μm),柱温为30 ℃,流速为0.5 mL?min-1。分离美托洛尔最佳条件为10 mmol?L-1磷酸盐缓冲液(pH值至5.0,含15 mmol?L-1 CM-β-CD)—甲醇(85∶15),柱温为30 ℃,流速为0.5 mL?min-1,检测波长为285 nm;分离文拉法辛最佳条件为10 mmol?L-1磷酸盐缓冲液(pH值至5.0,含20 mmol?L-1CM-β-CD)—甲醇(85∶15),柱温为 30 ℃,流速为0.5 mL?min-1,检测波长为230 nm。结果:在最佳分离条件下,美托洛尔、文拉法辛两种手性药物均达到了基线分离,分离度分别为1.51,1.79。结论:羧甲基-β-环糊精(CM-β-CD)对两种手性药物均有一定的手性选择性。

Abstract: Objective:To establish high performance liquid chromatography(HPLC) methods using carboxymethyl-β-cyclodextrin(CM-β-CD) as chiral mobile phase additives, and to study on enantiomeric separation of two kinds of chiral drugs methoprol and venlafaxine. Methods:The separation of metoprolol and venlafaxine was performed on the SunFireTM C18(4.6 mm×15 mm,5 μm) column.The column temperature was 30 ℃ and the flow rate was 0.5 mL?min-1. The optimal separation conditions of metoprolol was consisted of 10 mmol?L-1 phosphoric buffer (adjust the pH value of 5.0,consisted with 15 mmol?L-1 CM-β-CD-methanol(85∶15); The detection wavelength of metoprolol was 285 nm.The optimal separation conditions of venlafaxine was consisted of 10 mmol?L-1 phosphoric buffer(adjust the pH value of 5.0,consisted with 20 mmol?L-1 CM-β-CD)-methanol (85∶15); The detection wavelength of venlafaxine was 230 nm. Results: Under the optimal separation conditions, the enatiomers of metoprolol and venlafaxine were completely baseline separated, and their separation were respectively 1.51, 1.79.Conclusion: CM-β-CD has stereoselectivity for the two drugs.
