摘要: 【】目的:建立以HPLC-ELSD法测定单刀根多糖中葡萄糖含量的方法。方法 以葡萄糖为对照品,采用Prevail Carbohydrate ES(4.6?250mm,5μm)色谱柱,以水-乙腈 (25:75)为流动相,柱温:35℃,漂移管温度95℃,载气流量为2.5L?min-1。结果 葡萄糖线性关系良好,相关系数R=0.9991, 平均加样回收率为97.6%。结论 该方法简单、准确、重现性好,可用于单刀根药材多糖中葡萄糖含量的测定

Abstract: Objective The method to determination of glucose in polysaecharides of ethnodrug Millettia pulchra Kurz var.Laxio(r Dunn)Z.Wei. by HPLC- ELSD was establish . Method Using glucose as a standard,chromatography was performed on a wondasil Prevail Carbohydrate ES(4.6×250mm,5μm), the mobile phase was methyl alcoho and water(25:75), column temperature was 35 ℃, drift tube temperature was 95℃ ,Flow rate was 1.4 mL/min.Results Glucose had a perfect linear correlation, the correlation coefficient was 0.9991,the recovery was 97.6%.Conslusion The method is simple,accurate and reproducible,which can be used as glucose in polysaecharides of ethnodrug Millettia pulchra Kurz var.Laxio(r Dunn)Z.Wei. analysis.
