伊潘立酮包合物的制备及处方工艺优化

摘要: 目的:通过制备伊潘立酮-β-环糊精包合物,对其进行相应的验证,采用不同的制备方法对其制备工艺进行考察,以确定最佳工艺。方法:采用溶液-搅拌法制备包合物;以包合物的包合率为处方筛选出最佳工艺的依据;以包合温度(A)、包合时间(B)、伊潘立酮和β-CD投药比(包合比)(C)为考察因素;用溶出试验来对包合物进行相应的验证。结果:伊潘立酮的最佳工艺为伊潘立酮:β-CD(摩尔比)=1∶6,包合温度55 ℃,包合时间40 min。伊潘立酮包合物的形成导致伊潘立酮在水中的累计释放率有了明显增加。结论:证明了伊潘立酮包合物的制备工艺是可行的,通过改变加工工艺能够改变包合物的产量,包合物能够提高伊潘立酮的溶解性,为伊潘立酮开发出新的剂型提供参考依据。

Abstract: Objective: To investigate the preparation process of Iloperidone-β- cyclodextrin inclusion and to determine the best processing technology. Methods: The inclusion compound was prepared by solution-stirring method. According to the inclusion rate of the inclusion compound, the best technology was selected. The inclusion temperature (A), the time of inclusion (B), Iloperidone and β-CD were the factors to be investigated. The inclusion compound was verified by dissolution test. Results:The Iloperidone : The best technology for Iloperidone:β-CD (mole ratio)=1∶6, inclusion temperature of 55 ℃, closure time 40 min. The formation of the enalpinone inclusion complex resulted in a significant increase in the cumulative release rate of iparidone in water. Conclusion: Proved the Iloperidone inclusion compound preparation technology is feasible, by changing processing technology can change the yield of inclusion compound, inclusion compound can improve Iloperidone solubility, for Iloperidone provided a reference for develop new dosage forms.
