摘要: 目的:建立UPLC-MS/MS法测定多潘立酮的含量。方法:色谱条件为:色谱柱Agilent ZORBAX Eclipse Plus C18(2.1 mm×50 mm,5 μm),流动相A:0.1%甲酸溶液,流动相B:乙腈,流速0.3 mL·min-1,柱温40 ℃,进样量2 μL。质谱条件为:电喷雾离子化源,正离子监测模式(ESI+),雾化器压力:45 psi,干燥气流速:8.0 mL·min-1,干燥气温度:300 ℃,鞘气流速:10.0 mL·min-1,鞘气温度:325 ℃,气体为高纯氮气,离子源电压为5 500 V,扫描模式:MRM多反应监测扫描。结果:多潘立酮在1.012~10.12 μg·mL-1高浓度范围内线性关系良好,平均回收率为97.79%(RSD=1.5%);在0.506 0~50.60 μg·L-1低浓度范围内线性关系良好,平均回收率为95.73%(RSD=1.3%)。结论:本法快速、简便、专属性好、灵敏度高,可以为多潘立酮的质量标准研究提供理论依据。

Abstract: Objective: To establish an UPLC-MS/MS method for determining the content of domperidone. Methods:The determination was performed on a column Agilent ZORBAX Eclipse Plus C18(2.1 mm×50 mm,5 μm) with a mixture of 0.1% formic acid and acetonitrile as mobile phase,the flow rate was 0.3 mL·min-1, the column temperature was 40 ℃ and the injection volume was 2 μL. MS conditions: Electrospray ionization source, positive ion monitoring mode(ESI+), the nebulizer pressure was 45psi, the dry gas flow rate was 8.0 mL·min-1, the dry gas temperature was 300 ℃, the sheath gas flow rate was 10.0 mL·min-1, the sheath gas temperature was 325 ℃, the gas was high purity nitrogen, the ion source voltage was 5 500 V and the scan mode was MRM multi-reaction monitoring scan. Results:A good linearity was obtained in high concentration range of 1.012~10.12 μg· mL-1 of domperidone and the average recovery was 97.79%(RSD=1.5%), in addition, a good linearity was obtained in low concentration range of 0.506 0~50.60 μg·L-1 of domperidone and the average recovery was 95.73%(RSD=1.3%). Conclusion:The method was fast, simple, specific and sensitive.It can provide a theoretical basis for the research of quality standards of domperidone.
