HPLC法测定银香颗粒中大黄素、大黄酚及大黄素甲醚的含量

Determination of Emodin, Chrysophanol and Emodin Methyl Ether in Yinxiang Keli by HPLC

摘要: 目的:应用HPLC法,对银香颗粒中大黄素、大黄酚及大黄素甲醚的含量进行测定,为银香颗粒的质量控制提供依据。方法:采用HPLC法测定处方中大黄素、大黄酚及大黄素甲醚的含量,用Symmetry C18(4.6 mm×250 mm,5 μm)色谱柱,以甲醇-0.5%磷酸溶液(86∶14)为流动相,检测波长为430 nm。结果:大黄素在0.020 376~0.611 28 μg范围内呈良好的线性关系(r=1),平均回收率为99.85%,RSD为0.2%;大黄酚在0.076 884~2.306 519 μg范围内呈良好的线性关系(r=1),平均回收率为99.30%,RSD为0.5%;大黄素甲醚在0.019 531~0.585 921 μg范围内呈良好的线性关系(r=1),平均回收率为98.44%,RSD为0.9%。结论:现行标准中只测定大黄酸单一成分的含量,不能客观评价银香颗粒的质量,增加大黄素、大黄酚及大黄素甲醚含量的测定,能更好地控制该制剂的质量,可为该药的质量控制提供依据。

Abstract: Objective: The content of emodin, chrysophanol and emodin methyl ether in Yinxiang Keli was determined by HPLC, which provided a basis for controlling the quality of Yinxiang Keli. Methods: The content of Emodin、Chrysophanol and Rheochrysidin were determined by HPLC. The chromatographic conditions were: Symmetry C18(4.6 mm×250 mm, 5 μm) column, mobile phase being methanol and 0.5% phosphoric acid(86:14),and the detection wavelength at 430 nm. Results: Emodin showed a good linearity within the range of 0.020 376-0.611 28 μg, r=1, the average recovery was 99.85%, and RSD was 0.2%. Chrysophanol showed a good linearity within the range of 0.076 884-2.306 519 μg, r=1, the average recovery was 99.30%, and RSD was 0.5%. Rheochrysidin showed a good linearity within the range of 0.019 531-0.585 921 μg,r=1,the average recovery was 98.44%,and RSD was 0.9%. Conclusion: The quality of Yinxiang Keli can not be objectively evaluated by only determining the content of single component of rhein in the current standard. In this paper, the determination of emodin, chrysophanol and emodin methyl ether can better control the quality of the preparation and better guide clinical medication.
