摘要: 阿尔兹海默病(Alzheimer′s disease, AD)是一种常见的中枢神经系统退行性疾病,主要发病于65岁以上的老年人群,通常又被称为原发性老年痴呆症。临床上常表现为记忆功能障碍、认知功能缺失、语言功能下降等,严重影响人们的生活与健康。AD的发病机制尚不明确,其被学界广泛公认的学说有:Aβ沉积及毒性作用学说、Tau蛋白过度磷酸化假说、胆碱能功能缺损学说、兴奋性氨基酸毒性学说以及炎症反应学说等。目前仍未有效的药物来预防、改善或逆转AD的发生,因此研发新型、有效的AD治疗药物成为全球关注的热点。本文对阿尔兹海默病发病机制的最新研究进展及临床用药作一综述,为进一步研究AD提供数据参考。

Abstract: Alzheimer′s disease (AD) is a common central nervous system degenerative disease, mainly affected by the elderly over 65 years old. Often referred to as primary AD. Clinically, it often manifests as memory dysfunction, lack of cognitive function, and decreased language function, which affect people′s life and health. The mechanisms of AD are still unclear, and its doctrines recognized by a wide range of scholars around the world are:Aβ deposition and toxicity theory, Tau protein hyperphosphorylation theory, cholinergic function deficit theory, excitatory amino acid toxicity theory, inflammatory response theory, etc. There are still no effective drugs to prevent, improve or reverse the occurrence of AD. Therefore, the development of new and effective therapeutic drugs has become a global hot spot. In this paper, the latest research progress and clinical drug use of AD pathology are reviewed in detail, which provides a theoretical basis for further research on AD.
