摘要: 目的:建立超高效液相色谱-串联质谱(UPLC-MS/MS)法测定大鼠血浆中布洛芬的浓度。方法:色谱条件为:色谱柱Agilent ZORBAX Eclipse Plus C18(2.1 mm×50 mm,5 μm),流动相A为 0.01%甲酸-0.05%甲酸铵水溶液,流动相B为乙腈,流速0.3 mL·min-1,柱温40 ℃,进样量5 μL。质谱条件为:电喷雾负离子多反应监测模式,雾化器压力:40 psi,干燥气流速:6.0 mL·min-1,干燥气温度:150 ℃,鞘气流速:10.0 mL·min-1,鞘气温度:175 ℃,气体为高纯氮气,离子源电压为3 800 V。结果:布洛芬在20.477 1~2 047.71 ng·mL-1浓度范围内线性关系良好,平均回收率为115%。结论:本法快速、简便、专属性好、灵敏度高,可以为布洛芬的质量标准研究提供理论依据。

Abstract: Objective:To establish an UPLC-MS/MS method for determining the concentration of ibuprofen in rat plasma. Methods:The UPLC system was performed on a Agilent ZORBAX Eclipse Plus C18(2.1 mm×50 mm,5 μm)column with a mixture of 0.01% formic acid-0.05% ammonium formate aqueous solution and acetonitrile as mobile phase,the flow rate was 0.3mL·min-1, the column temperature was 40 ℃ and the injection volume was 5μL. The MS system was performed using multiple reaction monitoring(MRM) and electrospray ionization in negative mode, with the nebulizer pressure being 40 psi, the flow of drying gas being 6.0 mL·min-1, the temperature of drying gas being 150 ℃, the flow of sheath gas being 10.0 mL·min-1,the temperature of sheath gas being 175 ℃, the gas being high purity nitrogen, and the ion source voltage being 3 800 V. Results: A good linearity was obtained in concentration range of 20.477 1~2 047.71 ng·mL-1 of ibuprofen and the average recovery was 115%. Conclusion:The method was fast, simple, specific and sensitive. It can provide a theoretical basis for the research of quality standards of ibuprofen.
