孕期地塞米松暴露对大鼠生长发育的影响

摘要:

目的:探讨孕期地塞米松暴露对孕鼠和胎鼠心脏、肾脏、肝脏和肺脏质量的影响。方法:妊娠期SD大鼠分为对照组和地塞米松组,在孕9~20 d期间,对照组大鼠肌注0.9%的氯化钠注射液0.2 mL·d-1,地塞米松组大鼠肌注地塞米松磷酸钠注射液0.2 mL·kg-1·d-1。妊娠20 d麻醉后剖腹取胎,测量孕鼠和胎鼠的体质量、心脏、肝脏、肾脏和肺脏质量。结果:与对照组相比,地塞米松组孕鼠的体质量增长率在孕9~17 d均显著降低(P<0.05),其肝脏和肺脏的质量均显著降低(P<0.01);胎鼠的体质量、心脏、肾脏、肝脏和肺脏的质量均显著降低,胎鼠肺脏和肝脏的脏器系数显著降低(P <0.01)。结论:孕期地塞米松暴露不仅可影响孕鼠肝脏和肺脏的发育,也影响胎鼠心脏、肾脏、肝脏和肺脏的生长发育。

Abstract:

Objective:To investigate the effects of dexamethasone exposure during pregnancy on pregnant rats and fetal rats. Methods:SD rats during pregnancy were divided into control group and dexamethasone experimental group. During pregnancy 9-20 days, rats in the control group were intramuscularly injected with 0.9% sodium chloride injection 0.2 mL·d-1, and rats in the dexamethasone experimental group were intramuscularly injected with dexamethasone sodium phosphate injection 0.2 mL·kg-1·d-1. The fetuses were collected by laparotomy after anesthesia on the 20th day of pregnancy, and the body weights, heart, liver, kidney and lung weights of pregnant and fetuses were measured. Results:Compared with the control group, the body mass growth rates of pregnant rats in the dexamethasone experimental group were significantly reduced from 9 to 17 days of pregnancy(P<0.05), while the quality of their liver and lungs were significantly reduced(P<0.01).The body mass, heart, liver, kidney and lungs of fetal rats in the dexamethasone experimental group were significantly reduced, and the organ coefficients of the lungs and liver were also significantly reduced(P<0.01). Conclusion:Dexamethasone exposure during pregnancy can not only affect the development of liver and lung in pregnant rats, but also affect the growth and development of heart, kidney, liver and lung in fetal rats.
