Quality Evaluation of Spleen Aminopeptide Oral Lyophilized Powder Based on Characteristic Chromatogram Combined with Chemical Pattern Recognition

摘要: 目的:建立脾氨肽口服冻干粉特征图谱分析方法,并结合化学模式识别法评价其质量,为其科学鉴定及质量控制提供依据。方法:通过高效液相色谱法(HPLC)建立特征图谱,采用化学模式识别方法中的聚类分析、主成分分析对22批样品(S1~S22)进行分类,同时采用正交偏最小二乘判别分析识别脾氨肽口服冻干粉的关键性成分。结果:依据22批次样品建立了其特征图谱,确定了14个共有峰,并指认了其中4个成分肌苷、酪氨酸、色氨酸、胸腺素β4;特征图谱相似度为0.979~0.993;依据聚类分析及主成分分析将22批样品分为两类,采用正交偏最小二乘判别分析进一步筛选出胸腺素β4等7个质量差异标志性成分。结论:建立的脾氨肽口服冻干粉特征图谱准确度良好,稳定可靠,可为控制和评价其质量提供参考。

Abstract: Objective: To establish an analytical method for the characteristic chromatogram of spleen aminopeptide oral lyophilized powder and evaluate its quality using chemical pattern recognition, providing a basis for its scientific identification and quality control. Methods: The characteristic chromatogram was developed using liquid chromatography (HPLC), and 22 batches of samples (S1-S22) were classified using cluster analysis and principal component analysis from chemical pattern recognition methods. Additionally, orthogonal partial least squares-discriminant analysis was employed to identify key components distinguishing spleen aminopeptide oral lyophilized powder. Results: The characteristic chromatogram was established based on 22 batches of samples, identifying 14 common peaks and confirming four components: inosine, tyrosine, tryptophan, and thymosin β4. The samples were divided into two categories based on cluster analysis and principal component analysis. Orthogonal partial least squares discriminant analysis further screened seven quality-differentiating marker components, including thymosin β4. Conclusion: The characteristic chromatogram of spleen aminopeptide oral lyophilized powder established in this study is accurate, stable, and reliable, providing a reference for controlling and evaluating its quality.
