液质联用法检测延胡索饮片中5种违规添加着色剂

Quantitative Determination of Five Illegal Colorants in Corydalis Yanhusuo Decoction Pieces by HPLC-MS/MS

摘要: 目的:建立液质联用法检测延胡索饮片中5种人工着色剂(柠檬黄、日落黄、金橙Ⅱ、金胺O、酸性黄36)。方法:采用高效液相串联质谱(HPLC-MS/MS)法,色谱柱为Aglient Eclipse Plus C18(2.1 mm×100 mm,2.2 μm),流动相为乙腈-0.02 mol·L-1乙酸铵溶液(梯度洗脱),柱温40 ℃,流速0.3 mL·min-1,电喷雾(ESI)离子源,正、负离子扫描(MRM模式)。结果:柠檬黄、日落黄在0.40~20.00 μg·mL-1范围内,金橙Ⅱ、金胺O、酸性黄36在0.05~2.50 μg·mL-1范围内线性关系良好,线性相关系数r均大于0.99;日落黄、柠檬黄的检出限为0.1 μg·mL-1,金橙Ⅱ、金胺O、酸性黄36的检出限为0.02 μg·mL-1,平均回收率为 89.6% ~97.5%。采用该方法对市售的10批延胡索饮片进行检测,均未检出上述5种人工着色剂。结论:建立的方法具有高特异性和极低检测限,能快速检测延胡索饮片中是否含有非法人工着色剂,可用于延胡索饮片的质量控制。

Abstract: Objective: To establish a method for the determination of five yellow pigments (Lemon yellow, Sunset yellow, Orange Ⅱ, Auramine O, Acid yellow 36) added into corydalis rhizoma pieces by HPLC-MS/MS. Methods: The analysis was performed with Aglient triple quadrupole mass spectrometer.Electrospray ion(ESI)was adopted as the ion source with positive and negative ion scanning mode(MRM).The Aglient Eclipse Plus C18 (2.1 mm×100 mm, 2.2 μm) column was used with acetonitrile -0.02 mol·L-1 ammonium acetate solution as the mobile phase.The flow rate was 0.3 mL·min-1, the columm temperature was 40 ℃. Results: The result showed good linearity in the ranges of 0.40-20.00 μg·mL-1 for lemon yellow and sunset yellow, 0.05-2.50 μg·mL-1 for orange Ⅱ, auramine O and acid yellow 36. The limits of detection were 0.1 μg·mL-1 and 0.02 μg·mL-1.The average recovery of five yellow pigments was in the range of 89.6%-97.5%.None of the above five yellow pigments were detected in 10 batches of corydalis rhizoma pieces. Conclusion: The method is convenient, reliable and accurate, and can be used for the detection of yellow pigments in corydalis rhizoma pieces. It′s necessary for the quality control of corydalis rhizoma pieces.
