基于UPLC-Q-TOF/MS技术的 枳实大鼠血清药物化学研究

Study on Rat Serum Pharmacochemistry of Fructus Aurantii Immaturus Based on UPLC-Q-TOF/MS Method

摘要: 目的:运用血清药物化学方法分析大鼠口服枳实后的入血成分,初步阐明其可能的药效物质基础。方法:采用Agilent ZORBAX SB-C18(3.0 mm×100 mm,1.8 μm)色谱柱,以0.1%甲酸水(A)-乙腈(B)为流动相梯度洗脱,飞行时间质谱作为检测器分析样品。通过比较枳实提取物、空白血浆以及给药血浆的图谱差异,根据保留时间、精确相对分子质量、二级质谱裂解碎片及代谢产物的中性丢失等,鉴定大鼠灌胃给予枳实提取物后的血中原形成分及其代谢产物。结果:在给药后的大鼠血清中分析表征了21个入血成分,其中15个原形成分、6个代谢产物。结论:借助UPLC-Q-TOF/MS方法鉴定了大鼠口服枳实后血清中的化学成分,初步分析了枳实的入血成分,为阐明枳实质量标志物提供了理论依据。

Abstract: Objective: To analyze the blood components of rats after oral administration of the Fructus Aurantii Immaturus extract by the serum pharmacochemistry method to elucidated the effective constituents of Fructus Aurantii Immaturus. Methods: The samples were analyzed on Agilent ZORBAX SB-C18(3.0 mm×100 mm, 1.8 μm) column with 0.1% formic acid water(A)-acetonitrile(B) as mobile phase gradient elution and time-of-flight mass spectrometry as detector.. Based on the retention time, precise molecular weight, secondary mass spectrometry fragmentation and neutral loss of metabolites, compared with the difference of profiles of Fructus Aurantii Immaturus extract, blank serum and administered serum, and identified the prototypes and metabolites in blood of rats after administration of Fructus Aurantii Immaturus extract. Results: 21 components were analyzed and characterized in the rat serum after administration, including 15 prototypes and 6 metabolites. Conclusion: The chemical components in serum of Fructus Aurantii Immaturus were identified by UPLC-Q-TOF /MS method, the chemical components in the rats serum after oral administration of the Fructus Aurantii Immaturus extract were preliminarily identified, and it could provided theoretical basis for definition the Q-Marker of Fructus Aurantii Immaturus.
