问荆药材中异槲皮苷的定性和定量分析方法

Reach on the qualitative and quantitative analysis methods of Isoquercitrin in Equisetum arvens L.

摘要: 目的 不同生长环境及采收期对问荆药材异槲皮苷的影响,并建立定性和定量的分析方法 方法 采用薄层色谱法,对问荆药材中异槲皮苷进行定性分析;采用HPLC法对问荆药材中异槲皮苷进行含量测定, ZORBAX SB-C18(4.6mm×250mm,5μm)为色谱柱;流动相:乙腈-0.1%甲酸(15:85),流速为1.0mL/min;检测波长为354nm;进样量为10μL。结果 生长环境及采收期对薄层色谱和异槲皮苷含量均有较大的影响;薄层色谱斑点清晰,分离度良好;异槲皮苷在0.41μg/mL~95.13μg/mL范围内与峰面积呈良好线性,r=0.9999,平均回收率为97.2%,RSD=1.3%。结论 6月~8月是问荆药材的最佳采收期;分析方法简便,灵敏,重现性良好,可作为问荆药材的质量控制方法。

Abstract: Objective Study on different growth environment and harvest time on the quality of Equisetum arvens L .To establish methods for identification and determination of Isoquercitrin in Equisetum arvens L. Method TLC was used to identify Isoquercitrin in Equisetum arvens L, and the content was determine by HPLC, Agilent ZORBAX SB-C18(4.6mm×250mm,5μm) column was used. Acetonitrile and 0.1% Formic acid water-solution were used as mobile phase of liner elution, and the volume flow was at 1.0mL/min; Ultraviolent determination wavelength was 354nm; and injection volume was 10μL.Results Different growth environment and harvest time influence obviously TLC and the content of Isoquercitrin. TLC spots were clear, and sparated nicely. the linear range of Isoquercitrin was 0.41μg/mL~95.13μg/mL; The average recovery in Equisetum arvens L was 97.2%. Conclusion June and August are the best harvest time . The method is believable for identitication and determination of Isoquercitrin in Equisetum arvens L. with its simplicity, sensibility, repeatability。
