摘要： 目的:建立以高效液相色谱法测定科博肽含量的方法。方法: 色谱柱为Platisil ODS 5μ,150CcCc4.6mm,以0.1%三氟醋酸溶液为流动相A,三氟醋酸-50%乙腈(1:1000)为流动相B；柱温36℃；流速为每分钟1ml；检测波长为214nm；洗脱初始状态流动相B为8%,保持4分钟,在12分钟内,流动相B增至100%,保持6分钟,再在6分钟内回到初始状态,保持10分钟,进样量为：20μl。结果: 科博肽检测浓度的线性范围为：0.0775~0.62 mg.mL_-1,(r=0.9999),平均回收率为99.62%,RSD=1.63%。结论: 本方法操作简便、灵敏、准确、专属性强,可作为科博肽的含量测定方法。

Abstract: Objective：To develop a HPLC method for the determination of the content of Cobratide．Method：Cobratide was separated on Platisil ODS 5μ,150CcCc4.6mm, with 0.1% trifluoroacetic acid solution as the mobile phase A,trifluoroacetic acid - 50% acetonitrile (1：1000) as the mobile phase B；column temperature set at 36℃； Flow rate：1.0ml/min, the wavelength was set at 214nm；ElutionSof initial stateSB of the mobile phaseSwas 8%,hold for 4 minutes,within 12 minutes, increased to 100% mobile phase B,hold for 6 minutes,return to the initial state in 6 minutes,hold for 10 minutes,and the sample size was 20μl．Results：The standard curve of cobratide was linear with the range 0.0775~0.62 mg.mL_-1,(r=0.9999),with average recovery at 99.62%,RSD=1.63%．Conclusion：The method is simple,sensitive,accurate, specific,and can be used for the determination of Cobratide．
