摘要: 目的:建立五味子膏中葛根素含量的测定方法。方法:采用HPLC法测定葛根素含量,色谱柱为Diamonsil C18 (4.6 mm×250 mm,5 μm);甲醇-水(25∶75)为流动相;以250 nm作为检测波长;流速1.0 mL·min-1;检测器:DAD;柱温:30 ℃;进样量:10 μL。结果:葛根素40~1250 ng的范围内,线性关系良好(r=1);平均回收率为97.48%(RSD=1.2%)。结论:建立HPLC方法简单、重现性好、准确度高,可用于控制五味子膏中葛根素的含量。 [关键词] [中图分类号]

Abstract: Objective: To establish HPLC method for determination of puerarin in Wuweizigao. Methods: HPLC was used to determine the content of puerarin with Diamonsil C18(4.6 mm×250 mm,5 μm)column. The mobile phase was methanol-water(25∶75).The detection wavelength is 250 nm,the flow rate was 1.0 mL·min-1, and the column temperature was 30 ℃. Results: The puerarin linear ranges was 40-1250 ng.The linear equations of puerarin was in good linear range (r=1).The average recovery of Puerarin was 97.48% (RSD=1.2%). Conclusion: The method is simple, accurate and reproducible, which can be used for the quality control of Wuweizigao.
