摘要: 目的:建立快速、灵敏的LC-MS/MS法定量测定马血浆中的地塞米松浓度。方法:血浆样品经叔丁基甲基醚液液萃取后,使用ACE-C18色谱柱 (75mm×2.1mm, 3μm);采用ESI离子源MRM质谱扫描模式对血浆中的地塞米松浓度进行定量检测。结果:地塞米松血浆浓度测定方法的线性范围为0.01~1 0 ng.mL、日内/日间准确度分别为98.0%~120.0% 和104.0%~120.0%。日内/日间精密度分别为1.6~8.9%和2.2~9.7%。结论:该方法高效、准确、灵敏,可用于地塞米松在大动物模型中的药代动力学研究。

Abstract: ABSTRACT: OBJECTIVE To establish a rapid and sensitive method for quantification of dexamethasone in equine plasma using liquid chromatography-tandem mass spectrometry (LC-MS/MS). METHODS Analyte was recovered from plasma by liquid-liquid extraction using methyl tert-butyl ether (MTBE), separated on a C18 column, and analyzed in positive electrospray ionization mode. Multiple-reaction monitoring (MRM) was employed for quantification. RESULTS The linear dynamic range of the method was 0.01-10 ng·mL-1. The precision and accuracy at 0.05, 0.5 and 5 ng·mL?1 of dexamethasone during intra-day assay were 1.6-8.9% and 98-120%, respectively. For inter-day assay, precision and accuracy at the same three concentrations were 2.2-9.7% and 104-120%, respectively. CONCLUSION The present method is fast, simple, sensitive and reproducibly reliable, could be applied to the pharmacokinetics study of dexamethasone.
