Determination of Pyroglutamic Acid in human plasma by HPLC

摘要: 目的:建立HPLC法测定人血浆中焦谷氨酸含量的方法。方法:采用HPLC色谱法,色谱柱为 Agilent ZORBAX SB-Aq C18柱(4.6 mm×250 mm,5 μm),流动相为乙腈-辛烷磺酸钠溶液,梯度洗脱,流速为1.0 mL·min-1,检测波长为210nm,柱温35 ℃,进样体积20 μL。结果:焦谷氨酸在0.0025~0.02 mmol·L-1范围内线性关系良好(R=0.9998),加标回收率在97.2%~111.5%,RSD在0.7%~1.7%。结论:该法简便准确,可用于血浆中焦谷氨酸的测定。

Abstract: Objective: A HPLC method for the determination of Pyroglutamic Acid in human plasma was established. Methods: Chromatographic separation was performed in Agilent ZORBAX SB-Aq C18 column(4.6 mm×250 mm,5 μm)with an eluent consisting of acetonitrile and sodium 1-octanesulfonate solution at a flow rate of 1.0 ml.min-1. The eluent was monitored at 210 nm and the injection volume was 20 μL. Results: The linear range was 0.0025~0.02 mmol·L-1(r2=0.9997), and the recovery range was 97.2%~111.5% with RSD 0.7%~1.7%. Conclusion: The method was simple, accurate,which can be used for the determination of Pyroglutamic Acid in human plasma.
