Determination of Catechin Content in Testa Arachis in Different Areas by HPLC

摘要: 目的:建立花生衣药材中儿茶素含量测定方法,并与不同来源花生衣药材中儿茶素的含量进行比较。方法:采用HPLC法,以Sepax BR-C18(4.6 mm×250 mm 5 μm)为色谱柱;以乙腈-水(10∶90)为流动相,流速1.0 mL·min-1 ;柱温30 ℃;检测波长280 nm。结果:儿茶素在21.15~338.39 μg·mL-1范围内线性关系良好,相关系数为0.999 8;平均回收率为106%,RSD为1.7%。结论:该方法快速、准确,重复性好,为花生衣中儿茶素含量测定方法提供科学依据。实验表明,吉林省内花生衣药材中儿茶素含量较高。

Abstract: Objective: To establishe the method for the determination of catechin in testa arachis, and compare the catechin content in different sources of testa arachis. Methods: Using HPLC method, Sepax BR-C18 chromatographic column (4.6 mm×250 mm,5 μm) was used. Taking acetonitrile-water as the flowing phase, the flow velocity was 1.0ml, Column temperature was 30 ℃. Detection wavelength: 280 nm. Results: The interline relationship of catechin was good in the range of 21.15-338.39μg·mL-1, where the correlation coefficient was 0.999 8.The average recovery was 106% and RSD 1.7%. Conclusion: The method is fast, accurate and reproducible, which provides a scientific basis for the determination of catechin in testa arachis. The results showed that the content of catechin was higher in testa arachis in jilin province.
