胶塞中抗氧剂测定及其在药品中的迁移研究

Determination of Antioxidants in Rubber Plugs and Study on the Migration in Drug

摘要: 目的: 建立高效液相色谱(HPLC)法,测定注射用冷冻干燥用卤化丁基橡胶塞中抗氧剂BHT、1010、1076、330、168及其在注射用奥美拉唑钠中的迁移。方法:Agilent Zorbax Eclipse XDB-C18色谱柱(4.6 mm×250 mm,5 μm),以甲醇-水为流动相梯度洗脱,检测波长278 nm,流速1.0 mL·min-1,柱温30 ℃,进样量10 μL,结果:5种抗氧剂在相应浓度范围内线性关系良好(r≥0.999 1),在胶塞中回收率86.38%~102.37%,RSD 0.29%~1.85%,在药品迁移中回收率97.31%~100.04%,RSD 0.77%~1.91%。结论:该方法准确、灵敏,可用于胶塞中抗氧剂测定及其在药品中迁移研究。

Abstract: Objective: To establish high performance liquid chromatography (HPLC) to determine the antioxidants BHT, 1010, 1076, 330, 168 and their migration in omerazole sodium for injection in halogenated butyl rubber plug for freeze-drying for injection. Methods: Agilent Zorbax Eclipse XDB-C18 column (4.6 mm×250 mm,5 μm). Use methanol-water as mobile phase gradient elution, the detection wavelength was 278 nm and the flow rate was 1.0 mL·min-1. The column temperature was 30 ℃. And the injection volume was 10 μL. Results: The five antioxidants have a good linear relationship in the corresponding concentration range (r≥0.999 1). The recovery rate of the five kinds of antioxidants in the rubber stopper was 86.38%~102.37%, RSD 0.29%~1.85%. The recovery rate in drug migration was 97.31%~100.04%, RSD 0.77%~1.91%. Conclusion: This method is accurate and sensitive, and can be used for the determination of the antioxidants in rubber plug and migration studies in drugs.
