UPLC-DAD法测定款冬花中克氏千里光碱的含量

Determination of Senkirkine in Farfarae Flos by UPLC-DAD

摘要: 目的: 建立UPLC-DAD法测定款冬花中克氏千里光碱的含量。方法: 采用沃特世CORTECS UPLC C18(2.1 mm×100 mm, 1.6 μm)色谱柱,流动相为乙腈-0.1%磷酸水溶液(15∶85);检测波长220 nm;柱温:30 ℃;进样量:1 μL。结果: 克氏千里光碱在2.048~81.82 μg·mL-1浓度范围内与峰面积呈现良好的线性关系(r=1.000 0);平均加样回收率分别为89.46%,RSD分别为2.80%;10批样品中克氏千里光碱的含量为0.002 36~0.005 34%。结论:该方法简便、快速,可用于款冬花中克氏千里光碱的含量测定。

Abstract: Objective: To establish an UPLC-DAD method for quantitative analysis of senkirkine in Farfarae Flos. Methods: The assay was performed on an Waters CORTECS UPLC C18(2.1 mm×100 mm, 1.6 μm) column and the sample was eluted with a mobile phase consisted of acetonitrile -0.1 % phosphoric acid aqueous solution (15∶85). The detection wavelength was set at 220 nm, and the column temperature was 30 ℃. Injection volume: 1 μL. Results: The linear range of senkirkine was 2.048-81.82 μg·mL-1 (r>1.000 0). The average recovery was 89.46% with RSD of 2.80%. The contents of senkirkine in Farfarae Flos were 0.002 36-0.005 34%. Conclusion: The proposed method is simple and rapid, can be used as content determination of senkirkine in Farfarae Flos.
