HPLC法同时测定小儿麻甘颗粒中4种活性成分的含量

Simultaneous Determination of 4 Index Components in Xiaoermagan Granules by HPLC

摘要: 目的:建立HPLC法同时测定小儿麻甘颗粒中盐酸麻黄碱、苦杏仁苷、迷迭香酸和黄芩苷4个成分的含量。方法: 采用WondaSil C18-WR(4.6 mm×250 mm,5 μm)(W)色谱柱,以乙腈-0.2%磷酸溶液为流动相进行梯度洗脱,流速:1 mL·min-1,紫外检测波长为215 nm(0~6 min,测定盐酸麻黄碱)、207 nm(6~10 min,测定苦杏仁苷)、330 nm(10~24 min,测定迷迭香酸)、280 nm(24~30 min,测定黄芩苷)。结果: 4种成分色谱峰分离良好,盐酸麻黄碱、苦杏仁苷、迷迭香酸和黄芩苷分别在3.07~122.75、2.58~103.16、2.16~86.53、5.05~202.11μg·mL-1浓度内与峰面积具有较好线性关系;4种成分加标回收率为99.17%(RSD=1.35%)、98.74%(RSD=1.41%)、97.01%(RSD=1.77%)和99.45%(RSD=1.31%)。结论: 经过严格的试验确认,本方法操作简便、快速,结果准确可靠,可用于小儿麻甘颗粒中盐酸麻黄碱、苦杏仁苷、迷迭香酸和黄芩苷的含量测定。

Abstract: Objective: To develop an HPLC method for simultaneous determination of Ephedrine Hydrochloride,Amygdalin,Rosmarinic acid,Baicalin in Xiaoermagan Granules. Methods: The chromatographic separation was carried out on a WondaSil C18-WR (4.6 mm×250 mm,5 μm)(W) column with a mobile phase consisting of acetonitrile-2 mL·L-1 H3PO4 in a gradient mode at a flow rate of 1 mL·min-1. The UV detection wavelength was set at 215 nm for Ephedrine Hydrochloride in the first 6 min,207 nm for Amygdalin during 6-10 min, 330 nm for Rosmarinic acid during 10-24 min,280 nm for Baicalin during 24-30 min. Results: Excellent chromatographic separation was achieved and the ranges for linear correlation of Ephedrine Hydrochloride,Amygdalin,Rosmarinic acid,Baicalin were 3.07-122.75、 2.58-103.16、2.16-86.53 and 5.05-202.11μg·mL-1, respectively. The average recovery of the four components were 99.17% (RSD=1.35%)、98.74% (RSD=1.41%)、97.01% (RSD=1.77%) and 99.45% (RSD=1.31%). Conclusion: The established method is simple,rapid,sensitive and accurate, which can be applied to determine Ephedrine Hydrochloride,Amygdalin,Rosmarinic acid,Baicalin in Xiaoermagan Granules.
