亚抑菌浓度亚胺培南对多重耐药鲍曼不动杆菌生物膜作用的研究

Study on the Effect of Imipenem at Subinhibitory Concentration for the Biofilm of Multi-drug Resistant Acinetobacter Baumannii

摘要: 目的:探讨亚抑菌浓度亚胺培南对多重耐药鲍曼不动杆菌生物膜的影响。方法: 采用微量肉汤稀释法,测定医院2019年2月至2020年9月临床分离的36株亚胺培南耐药多重耐药鲍曼不动杆菌(MDR-AB)的最低抑菌浓度(MIC),结晶紫半定量染色法测定菌株生物膜形成水平。采用96孔聚苯乙烯板体外构建鲍曼不动杆菌生物膜的模型,用亚抑菌浓度亚胺培南作用于成熟生物膜。实验组Ⅰ、Ⅱ、Ⅲ组中分别用MIC、1/2MIC、1/4MIC的亚胺培南作用于已形成的成熟生物膜;对照组Ⅳ组不加亚胺培南。通过结晶紫半定量染色法对生物膜进行染色,共聚焦倒置显微镜下观察生物膜染色情况,采用多功能酶标仪在570 nm波长处测得生物膜OD值。结果: 亚胺培南在实验组Ⅰ、Ⅱ、Ⅲ组中,即 MIC、1/2MIC、1/4MIC 浓度下作用于已成熟生物膜 48、96 h后,所测得OD值与对照组 Ⅳ组OD值之间进行比较,有显著性差异(P<0.05)。结论: 亚抑菌浓度亚胺培南具有破坏成熟生物膜的能力,且随着浓度的升高,其破坏能力更强。

Abstract: Objective: To investigate the influnce of Imipenem in Sub-inhibitory concentration against the biofilm formation of Multiple drug resistant Acinetobacter Bauman (MDR-Ab). Methods: The minimum inhibitory concentration (MIC) of imipenem against 36 imipenem-resistant multidrug-resistant Acinetobacter baumannii (MDR-Ab) strains isolated from a third-class A hospital from February 2019 to September 2020 was determined by microbroth dilution method. The biofilm formation level of the strains was determined by semi-quantitative crystal purple method. The biofilm model of Acinetobacter baumannii was constructed by 96-well polystyrene plate in vitro. After the mature biofilm was formed, imipenem at subinhibitory concentration was applied to the mature biofilm. The mature biofilms in experimental groups Ⅰ,Ⅱ and Ⅲ were treated with imipenem of MIC, 1/2MIC and 1/4MIC, respectively. Control group Ⅳ did not receive imipenem. The biofilm was stained by crystalline violet semi-quantitative staining, and the biofilm staining was observed under a confocal inverted microscope. The OD value of the biofilm was measured at 570 nm by a multifunctional enzyme plate analyzer. Results: In experimental groups Ⅰ,Ⅱ and Ⅲ, i.e. MIC, 1/2MIC and 1/4MIC, imipenem was applied to the mature biofilm for 48h and 96h, the measured OD values were compared with the control group, and there were significant differences (P<0.05). Conclusion: The subinhibitory concentration of imipenem has the ability to destroy mature biofilm, and the damage ability becomes stronger with the increase of the concentration.
