HS-GC法测定枸地氯雷他定原料药中12种有机溶剂的残留量

Determination of 12 Residual Organic Solvents in the Raw Material of Desloratadine Citrate Disodium by HS-GC

摘要: 目的:建立同时测定不同合成工艺的枸地氯雷他定原料药中12种有机溶剂残留量的方法。方法:采用顶空气相色谱(HS-GC)法。色谱柱为Agilent DB-624 (30 m×0.25 mm,1.4 μm)毛细管柱,程序升温,进样口温度为250 ℃,检测器为氢火焰离子化检测器,检测器温度为300 ℃,载气为高纯氮气,载气流速为1.0 mL·min-1,分流比为50∶1,顶空进样量为1 mL,顶空平衡温度为90 ℃,顶空平衡时间为30 min。结果:12种有机溶剂在建立的色谱条件下均能完全分离,考察浓度范围内质量浓度与峰面积呈良好线性关系,精密度和准确度结果良好。结论:该方法操作简单、灵敏、结果准确,可用于不同合成工艺的枸地氯雷他定原料药中有机溶剂残留量的控制。

Abstract: Objective: To establish a method for determination of 12 residual organic solvents in desloratadine citrate disodium raw materials by different synthetic processes. Methods: Headspace gas chromatography(HS-GC) was adopted. The determination was performed on Agilent DB-624 (30 m×0.25 mm, 1.4 μm) capillary column using the temperature programming. The temperature of injector port was 250 ℃, and the FID detector was used with the temperature of 300 ℃. The carrier gas was nitrogen and the flow rate was 1.0 mL·min-1.The spilt ratio was 50:1, and the headspace injection volume was l.0 mL. The headspace equilibrium temperature was 90 ℃, and the equilibrium time was 30 min. Results: All the compounds were separated completely. The calibration curve of each compounds showed a good linear relationship with good precision and accuracy. Conclusion: The method is simple, sensitive and accurate, which can be applied on the organic solvent residue control of desloratadine citrate disodium raw material in different synthetic processes.
