复方氟尿嘧啶口服溶液中氟尿嘧啶含量测定方法的改进

Improvement on the Determination Method of Fluorouracil in Compound Fluorouracil Oral Solution

摘要: 目的:改进复方氟尿嘧啶口服溶液中氟尿嘧啶含量测定方法。方法:采用高效液相色谱法,色谱柱为Agilent 5 TC-C18(2)(4.6×250 mm,5 μm),流动相为6.8 g·L-1的磷酸二氢钾溶液(用5 mol·L-1氢氧化钾溶液调节pH值至5.7±0.1):甲醇(95∶5),检测波长为265 nm。结果:氟尿嘧啶在10.289 μg·mL-1~51.443 μg·mL-1浓度范围内线性关系良好,线性方程:Y=34.554X+5.479,(r=0.999 98),平均回收率为96%(RSD=0.9%,n=9)。结论:本文所建立高效液相色谱法简便、准确、灵敏度高、重复性好,满足复方氟尿嘧啶口服溶液中氟尿嘧啶的含量测定。

Abstract: Objective:To improve the determination method of fluorouracil in compound fluorouracil oral solution.Methods:HPLC was performed on the column of Agilent 5 TC-C18(2) with mobile phase of 6.8 g·L-1 potassium dihydrogen phosphate solution (adjust pH to 5.7 ± 0.1 with 5 M potassium hydroxide solution) - methanol (95∶5), detection wavelength was 265 nm. Results: The linear range of fluorouracil was 10.289 μg·mL-1-51.443 μg·mL-1. The linear equation was Y=34.554X+ 5.479(r=0.999 98). The average recovery was 96% (RSD=0.9%, n=9).Conclusion:The method is simple, accurate with high sensitivity and good reproducibility, and be used for the determination of fluorouracil in compound fluorouracil oral solution.
