不同产地中药桔梗中5种桔梗皂苷含量的测定

Determination of the Contents of Five Platycosides in Platycodonis Radix from Different Habitats

摘要: 目的:建立超高效液相色谱-电雾式检测器(UPLC-CAD,ultra performance liquid chromatography-charged aerosol detection)联用法同时测定桔梗中5种桔梗皂苷的含量,比较不同产地桔梗药材与饮片的质量差异。方法:采用Dikma Endeavorsil C18(2.1 mm×100 mm,1.8 μm)色谱柱,柱温为30 ℃,乙腈-水为流动相,梯度洗脱,流速0.3 mL·min-1,进样量2 μL,CAD检测器参数为雾化器温度70 ℃,载气(N2)压力60 psi。结果:去芹糖桔梗皂苷E、桔梗皂苷E、桔梗皂苷D3、去芹糖桔梗皂苷D和桔梗皂苷D分别在18.5~924.8 ng、20.5~1 025.2 ng、20.5~1 023.3 ng、23.3~1 163.4 ng和19.7~983.0 ng进样量范围内和峰面积线性关系良好,平均加样回收率分别为98.4%、100.6%、100.9%、103.9%和103.7%,RSD分别为1.0%、0.8%、1.6%、1.8%和1.4%。不同产地桔梗药材及饮片中桔梗皂苷D的含量均高于0.10%,其中安徽、山东等华东省份的桔梗中桔梗皂苷D含量均值达到0.20%以上,质量较佳;甘肃产桔梗中5种桔梗皂苷的含量均达0.10%以上,整体质量最优;桔梗药材中桔梗皂苷的含量略高于饮片;除桔梗皂苷D外,全部样品中桔梗皂苷E的含量亦均达0.10%以上,建议可增设为中药桔梗的质量评价指标。结论:不同产地的桔梗药材及饮片中5种桔梗皂苷的含量差异较大,本研究方法简便快捷,测定结果准确可靠,可为完善中药桔梗质量控制提供参考。

Abstract: Objective:To establish a aultra performance liquid chromatography-charged aerosol detection(UPLC-CAD) method for the simultaneous determination of five platycosides in Platycodonis Radix, compare the quality differences of samples from different habitats, and improve the current quality standard. Methods:The chromatographic conditions were Dikma Endeavorsil C18 column(2.1 mm×100 mm,1.8 μm),column temperature of 30 ℃, acetonitrile-water as mobile phase with gradient elution, flow rate of 0.3 mL·min-1. The parameters of CAD were nebulizer temperature of 70 ℃ and carrier gas(N2) pressure of 60 psi. Results: The linearity, precision, stability, reproducibility and average recovery rates of the five platycosides(deapio-platycodin E, platycodin E, platycodin D3,deapio-platycodin D and platycodin D) were good. The contents of platycodin D in all samples were more than 0.10%, especially the average values of samples from Anhui, Shandong reached 0.20%. The contents of the five platycosides in samples from Gansu were more than 0.10% and the quality was the best. The assay results showed that the contents of five platycosides in Platycodonis Radix were slightly higher than platycodon cut crude drug.In addition, the contents of platycodin E in all samples were more than 0.10%, which could be used as a new quality evaluation index of Platycodonis Radix. Conclusion:The contents of five platycosides in Platycodonis Radix from different habitats are quite different. The established method is simple, accurate and rapid, which can be used to improve the quality control of Platycodonis Radix.
