HPLC法测定川贝止咳露中防腐剂的含量

Determination of Preservatives in Chuanbei Cough Syrups by HPLC

摘要: 目的:建立HPLC法测定川贝止咳露中5种防腐剂含量的方法。方法:采用CAPCELL PAK-C18色谱柱(250 mm×4.6 mm,5 μm),以乙腈-0.2%磷酸水溶液为流动相,梯度洗脱;流速1.0 mL·min-1;柱温20 ℃;检测波长为230 nm(苯甲酸)和255 nm(山梨酸、羟苯甲酯、羟苯乙酯、羟苯丙酯);进样量10 μL。结果:43批药品中均可检出苯甲酸,33批药品中检出羟苯乙酯,2种防腐剂的检出量均在《中国药典》规定范围之内;所检药品批次中均未检测出山梨酸、羟苯甲酯和羟苯丙酯。结论:该方法操作简便,专属性强,可用于川贝止咳露中防腐剂的含量测定。

Abstract: Objective:To establish HPLC method for quantitative analysis of preservatives in Chuanbei Cough Syrups.Methods:The CAPCELL PAK-C18 chromatographic column (250 mm×4.6 mm,5 μm) was used for this test. Gradient elution was carried out with acetonitrile-0.2% phosphoric acid aqueous solution as the mobile phase at a flow rate of 1.0 mL·min-1, in which the column temperature is 20 ℃, and the detection wavelength are 230 nm(Benzoic acid) and 255 nm(Sorbic acid, Methyl hydroxybenzoate,Ethyl hydroxybenzoate and Propyl hydroxybenzoate).The injection volume is 10μL. Results: Benzoic acid was detected in 43 batches of drugs, Ethyl hydroxybenzene was detected in 33 batches of drugs, and the detection amount of the two preservatives was within the scope specified in Chinese Pharmacopoeia.Sorbic acid, Methyl hydroxybenzoate and Propyl hydroxybenzoate were not detected in the tested drug batches. Conclusion:The method is simple and specific, and can be used for the determination of preservatives in Chuanbei cough Syrups.
