Excel残数法计算房室模型PK参数

摘要: 目的:Excel残数法计算不同房室模型、不同给药途径的药物动力学(Pharmacokinetics,PK)参数,并与Phoenix WinNonlin、GastroPlus、PK solver统计软件的计算结果进行对比。方法:采用Excel残数法计算一房室静脉注射、血管外给药和二房室静脉注射、血管外给药药物的PK参数,并与Phoenix WinNonlin、PK solver和GastroPlus统计软件计算所得结果进行比较,评估各软件计算PK参数的一致性及准确性。结果:计算一房室静脉注射给药、血管外给药和二房室静脉注射给药、血管外给药的药代动力学参数,Excel残数法与其他3个统计软件计算结果无显著差异,4种评价方法准确性及一致性均较高。结论:残数法可准确计算房室模型药物的PK参数,且与Phoenix WinNonlin、PK solver、GastroPlus统计软件计算结果一致。

Abstract: Objective:The pharmacokinetic parameters of different atrioventricular models and different routes of administration were calculated by Excel residue method, and the results were compared with those calculated by different statistical software. Methods:Excel residue method was used to calculate the PK parameters of one-compartment intravenous injection, extravascular drug administration and two-compartment intravenous injection and extravascular drug administration, and the results were compared with those calculated by Phoenix WinNonlin, PK solver and GastroPlus statistical software. Results:Calculate the pharmacokinetic parameters of one-compartment intravenous administration, extravascular administration, two-compartment intravenous administration and extravascular administration. There is no significant difference between the results calculated by Excel residue method and other three statistical software,and four evaluation methods have high accuracy and consistency.Conclusion:The residue method can accurately calculate the PK parameters of atrioventricular model drugs, which is consistent with the results calculated by Phoenix WinNonlin, PK solver and GastroPlus statistical software.
