建立超高效液相色谱法测定莫匹罗星软膏含量

Establishment of Ultra Performance Liquid Chromatography for the Determination of Mupirocin Ointment

摘要: 目的:建立莫匹罗星软膏含量测定超高效液相色谱法,提高检测效率。方法:采用超高效液相色谱法(Ultra performance liquid chromatography,UPLC),色谱柱为ACQUITY UPLC BEH C18 (2.1 mm×100 mm,1.7 μm);流动相为0.01 mol·L-1磷酸二氢钠(1 mol·L-1 NaOH调节pH至6.3)-乙腈(75∶25);检测波长为230 nm;流速为0.4 mL·min-1。结果:建立了莫匹罗星软膏含量测定超高效液相色谱方法,此方法精密度好,准确度高,其线性范围为0.01~1 mg·mL-1,R2为0.999 6。结论:相比于高效液相色谱(High Performance Liquid Chromatography,HPLC)法,本文所提方法更便捷高效,可有效地控制莫匹罗星软膏含量。

Abstract: Objective:The aim of this study is to establish an ultra performance liquid chromatography method for the determination of mupirocin ointment and to improve the detection efficiency. Methods: In this study, ultra performance liquid chromatography (UPLC) method was used with the ACQUITY UPLC BEH C18 (2.1 mm×100 mm, 1.7 μm) as the chromatographic column. In this process, the mobile phase, detection wavelength and flow rate were 0.01 mol·L-1 sodium dihydrogen phosphate (adjusted pH to 6.3 by 1 mol·L-1 NaOH) -acetonitrile (75∶25), 230 nm and 0.4 mL·min-1, respectively. Results: An ultra performance liquid chromatography method with good precision and high accuracy for the determination of mupirocin ointment content was established with the linear range was 0.01-1 mg·mL-1 and the R2 was 0.999 6. Conclusion: Compared with High Performance Liquid Chromatography (HPLC), the proposed method is more convenient and efficient, and can effectively control the content of mupirocin ointment.
