柑橘类药材中辛弗林含量研究

Study on the Content of Synephrine in Citrus spp.

摘要: 目的:比较柑橘类药材(陈皮、青皮、枳实、香橼等)中辛弗林含量差异,为相关药材质量评价提供参考。方法:建立了柑橘类药材中辛弗林HPLC含量测定方法;分析采用Waters SPHERISORBSCX(4.6 mm×250 mm,5 μm)色谱柱,以甲醇-0.2%磷酸溶液(氨试液调pH至3.8)(35∶65)为流动相,检测波长223 nm,柱温30 ℃。比较了陈皮、青皮、枳实、香橼等柑橘类药材及陈皮不同栽培品种果皮中辛弗林的含量分布情况。结果:辛弗林在进样浓度2.842 8~568.560 6 μg·mL-1范围内线性关系良好(r=1.000 0);平均回收率为102.92%,RSD为1.39%(n=9)。柑橘类药材中辛弗林含量高低为青皮>枳实>陈皮(大红袍)>陈皮(茶枝柑)>橙皮>柠檬>枸橘,佛手、香橼、化橘红、柚皮、金橘中未检出。陈皮类药材不同栽培品种果皮中,大红袍陈皮中辛弗林含量最高,杂陈皮椪柑次之,杂陈皮不知火、春香中辛弗林含量最低。结论:柑橘类药材中辛弗林分布规律与其植物分类存在一定相关性,宽皮柑橘、橙这两类柑橘属药材中辛弗林含量较高,且未成熟果皮中辛弗林含量高于成熟果皮;而陈皮类药材不同栽培品种果皮中辛弗林含量也存在差异。本研究可为柑橘类药材的质量评价和应用提供一定参考和依据。

Abstract: Objective: To compare the content of synephrine in Citrus spp. (Pericarpium citri reticulatae, Pericarpium citri reticulatae viride,Aurantii fructus immaturus, etc.), and provide reference for the quality evaluation of related medicinal materials. Methods:HPLC method for determination of synephrine in Citrus spp. was established. Using Waters SPHERISORB SCX (4.6 mm×250 mm,5 μm) with a mobile phase of methanol-0.2% phosphoric acid solution (adjusted pH to 3.8 with ammonia test solution) (35∶65). The detection wavelength was 223 nm, and the column temperature was 30 ℃. The content of synephrine in Citrus spp. and different cultivars of Pericarpium citri reticulatae were compared. Results:The linear range of synephrine was 2.842 8-568.560 6 μg·mL-1(r=1.000 0). The average recovery was 102.92%(RSD=1.39%, n=9). The content of synephrine in Citrus spp. were Qingpi> Zhishi>Dahongpao>Chazhigan>Chengpi>Ningmeng>Gouju. Synephrine was not detected in Foshou, Xiangyuan, Huajuhong, Youpi and Jinju. Among the different cultivars of Pericarpium citri reticulatae, Dahongpao had the highest synephrine content, followed by Ponkan, and Buzhihuo and Chunxiang had the lowest synephrine content. Conclusion:There was a certain correlation between the distribution of synephrine in Citrus spp. and its plant classification. The content of synephrine in Sinocitrus Tseng and Aurantium Miller were higher, and the content of synephrine in immature peel was higher than that in mature peel. The content of synephrine were also different between cultivars of Pericarpium citri reticulatae. This study could provide a certain reference and basis for the quality evaluation and application of Citrus spp..
