九味羌活丸UPLC指纹图谱的建立及化学模式识别

Establishment of UPLC Fingerprint Chromatograms for JiuweiQianghuo Pills and Chemical Pattern Recognition

摘要: 目的:建立九味羌活丸的超高效液相色谱(UPLC)指纹特征图谱并开展化学模式识别。方法:采用Waters Acquity UPLC HSS T3(2.1 mm× 100 mm,1.8 μm)色谱柱,以0.1%磷酸-乙腈进行梯度洗脱,进样量1 μL,柱温为40℃,流速为0.35 mL·min-1,检测波长为254 nm。通过中药指纹图谱相似度分析软件及化学模式识别技术对12批九味羌活丸样品进行分析。结果:通过比对12批样品,识别出24个共有峰,建立九味羌活丸的UPLC指纹对照图谱并指认出其中13个成分;12批样品相似度均在0.90以上。聚类分析和主成分分析将12批样品分为3 类,通过正交偏最小二乘法-判别分析推测其中2个成分是引起批间差异的关键因素,可作为本品的质量标志性组分。结论:建立的九味羌活丸UPLC指纹图谱能较为全面地评价本品的内在质量,同时结合化学模式识别分析可为九味羌活丸的质量标准制定提供参考依据。

Abstract: Objective: Toestablish UPLC (Ultra-Performance Liquid Chromatography) fingerprint chromatograms for JiuweiQianghuo Pills and conduct chemical pattern recognition.Methods:Waters Acquity UPLC HSS T3 column (2.1 mm × 100 mm, 1.8 μm) was used. Gradient elution was performed with 0.1% phosphoric acid and acetonitrile. The injection volume was 1 μL, the column temperature was set at 40°C, the flow rate was 0.35 mL·min-1, and detection was carried out at 254 nm. Twelve batches of JiuweiQianghuo Pills were analyzed using traditional Chinese medicine fingerprint similarity analysis software and chemical pattern recognition techniques.Results:By comparing the 12 batches of samples, 24 common peaks were identified. UPLC fingerprint chromatograms of JiuweiQianghuo Pills were established, and 13 components were identified. The similarity values among the 12 batches of samples were all above 0.90.The 12 batches of samples were divided into 3 categories by cluster analysis and principal component analysis. The orthogonal partial least square method and discriminant analysis were used to predict that 2 of them were the key factors causing the difference between batches and could be used as the quality signature components of this product.Conclusion:The established UPLC fingerprint chromatograms for JiuweiQianghuo Pills can comprehensively evaluate the intrinsic quality of the product. Combined with chemical pattern recognition analysis, they can provide reference data for setting quality standards for JiuweiQianghuo Pills.
