基于网络药理学探讨温阳散寒活血方治疗狭窄性腱鞘炎的作用机理

Mechanism of Wenyang Sanhan Decoction in Treating Stenosing Tenosynovitis Based on Network Pharmacology

摘要: 目的:运用网络药理学方法探讨温阳散寒活血方治疗狭窄性腱鞘炎的作用机制。方法:运用中药系统药理学数据库与分析平台(TCSMP)检索筛选温阳散寒活血方的药物活性成分及潜在靶点。结合GeneCards和OMIM数据库搜索腱鞘炎的相关基因靶点。基于Uniprot数据平台对靶点的基因名称进行整理与规范,在STRING 11.5数据平台上生成温阳散寒活血方治疗腱鞘炎的靶点蛋白互作网络,利用Cytoscape 3.9.1软件对温阳散寒活血方治疗腱鞘炎的成分、作用靶点进行分析与网络的构建,利用DAVID 6.8平台对靶点进行基因本体(GO)功能富集与京都基因与基因组百科全书(KEGG)通路分析。 结果:最终筛选到温阳散寒活血方有效化学成分共计41个,作用靶点含有96个。温阳散寒活血方治疗腱鞘炎的核心活性成分有β-谷甾醇(β-sitosterol)、槲皮素(quercetin)、山柰酚(kaempferol)和杨梅酮(Myricanone)等,作用的关键靶点有IL6、VEGFA等。GO功能富集分析得到48个分子功能,KEGG富集分析得到47条信号通路,其中关系最密切的为TNF信号通路。结论:温阳散寒活血方通过多靶点、多途径抑制炎症反应,从而达到治疗腱鞘炎的目的。

Abstract: Objective: To explore the therapeutic mechanism of Wenyang Sanhan Decoction on stenosing tenosynovitis using method of network pharmacology. Methods:The active components and potential target genes of Wenyang Sanhan Decoction were searched and screened by Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform (TCMSP). These genes were mapped with the tenosynovitis related genes obtained by Genecards and OMIM database. The gene names of the targets were sorted out and standardized based on the Uniprot data platform, and String 11.5 database and Cytoscape 3.9.1 software were used to construct the protein-protein interaction(PPI) network and the active ingredient-core target network. Then, gene ontology (GO) and Kyoto Encyclopedia of Gene and Genome (KEGG) enrichment analysis of core target genes were carried out by using DAVID 6.8 platform. Results:A total of 41 effective chemical components and 96 action targets were screened out. The core active components of Wenyang Sanhan Decoction in the treatment of tenosynovitis are β-sitosterol, quercetin, kaempferol, myricetin and so on, and core target genes were mainly involved in IL6, VEGFA and so on. 48 molecular functions were obtained by GO functional enrichment analysis, and 47 signal pathways were obtained by KEGG enrichment analysis, of which TNF signal pathway was the most closely related. Conclusion:Wenyang Sanhan Decoction can inhibit inflammation through multiple targets and pathways, so as to achieve the purpose of treating tenosynovitis.
