舒脑欣滴丸对反复缺血再灌注所致小鼠记忆障碍模型的保护作用及其机制研究

Study on the Protective Effect and Mechanism of Shunaoxin Dropping Pill on Memory Impairment Model of Mice Induced by Repeated Ischemia-reperfusion

摘要: 目的:观察舒脑欣滴丸对小鼠反复脑缺血再灌注所致记忆障碍模型的保护作用,并对其作用机制进行探讨。方法:采用反复缺血再灌注制备小鼠记忆功能障碍模型,通过跳台法测定动物学习记忆障碍,通过全脑SOD、MDA含量测定,观察氧化应激因子变化;通过脑组织HE染色,观察脑组织形态学和海马神经元密度变化。结果:反复脑缺血引起的记忆障碍模型小鼠,用舒脑欣滴丸治疗能明显改善学习记忆障碍,学习后记忆阶段错误次数明显减少,潜伏期明显增加,氧化应激指标显著改善,海马CA1区神经元凋亡明显减少。结论:舒脑欣滴丸口服给药,对脑缺血再灌注所致记忆障碍模型小鼠有良好的治疗作用,其机制可能与抑制氧化应激和减少海马区神经元损伤有关。

Abstract: Objective: To observe the protective effect of shunaoxin dropping pill on the model of memory impairment induced by repeated cerebral ischemia-reperfusion in mice, and to explore its mechanism. Methods: The mouse model of memory dysfunction was made by repeated ischemia-reperfusion, and the learning and memory disorders were measured by the step-down method. The changes of oxidative stress factors were observed by the determination of SOD and MDA content in the whole brain. The brain morphology and the density of hippocampal neurons were observed by HE staining. Results: In the model mice of memory impairment caused by repeated cerebral ischemia, shunaoxin dropping pill could significantly improve the learning and memory impairment. The number of errors in the memory stage was significantly reduced, and the incubation period was significantly increased. The indexes of oxidative stress were significantly improved, and the apoptosis of hippocampal CA1 neurons was significantly reduced. Conclusion: Shunaoxin dropping pill has a good therapeutic effect on memory impairment model mice induced by cerebral ischemia-reperfusion, and its mechanism may be related to the inhibition of oxidative stress and reduction of hippocampal neuron damage.
