HPLC-ELSD法测定益气补血片中黄芪甲苷的含量

摘要: 目的:建立高效液相色谱-蒸发光散射检测法测定益气补血片中黄芪甲苷含量,用于指导该药的质量控制。方法:采用Sepax BR-C18色谱柱 (4.6 mm×250 mm, 5 μm),乙腈-水(35∶65)为流动相;流速1.0 mL·min-1;蒸发光散射检测器;柱温30 ℃。结果:黄芪甲苷质量浓度分别在1.02~10.27 μg·mL-1 (r=0.999)范围内线性关系良好,方法的平均回收率(n=6)分别为96.99%(RSD=2.00%);3批益气补血片中黄芪甲苷含量为0.101 mg/片~0.157 mg/片。同时采用原标准中的薄层扫描法进行测定,两种方法结果基本一致。结论:建立的黄芪甲苷含量测定方法可用于益气补血片的质量控制。

Abstract: Objective: To establish an HPLC-ELSD for the determination of astragaloside Ⅳ in Yiqibuxue Tablets, so as to instruct the quality control of the Chinese patent medicine. Methods: HPLC analysis was carried out a Sepax BR-C18 column (4.6 mm×250 mm, 5 μm). The mobile phase was acetonitrile-water (35∶65) with isocratic elution at the flow rate of 1 mL·min-1, the detector was an evaporative light-scattering detector (ELSD), the column temperature was 30 ℃. Results: The linear range of astragaloside Ⅳ was1.02-10.27 μg·mL-1 (r=0.999). The average recoveries (n=6) of astragaloside Ⅳ was 96.99% (RSD=2.00%). The contents of the medicine in 3 samples were 0.101 mg -0.157 mg in a tablet. These results were in good agreement with the ones obtained with the TLC scanning method of current quality standard. Conclusion: The established method for determination of astragaloside Ⅳ can be used for the quality control of Yiqibuxue tablets.
