诱变剂二甲肼对PC12细胞毒性效应及其机制研究

Study on the Toxic Effect of the Dimethylhydrazine on PC12 Cells and its Mechanism

摘要: 目的:研究二甲肼对PC12细胞的毒性作用及其作用机制。方法: 二甲肼分别以600、700、800、900、1 000 μmol·L-1浓度为作用剂量,与PC12细胞孵育48 h后,采用CCK-8比色法检测二甲肼对体外培养PC12细胞增殖的抑制作用;彗星实验技术检测PC12细胞DNA的损伤作用;流式细胞术考察细胞凋亡。结果: 二甲肼作用PC12细胞48 h后对其具有明显的毒性作用,细胞生长受到明显的抑制;单细胞凝胶电泳实验表明,二甲肼对体外培养PC12细胞的DNA有明显的损伤作用;流式细胞结果显示,二甲肼可诱导细胞凋亡。以上作用均具有浓度依赖性,且具有显著性差异(P<0.05)。结论: 二甲肼能够显著抑制PC12细胞增殖,损伤细胞DNA,并具有剂量依赖性,其作用机制可能和细胞内过氧化有关。

Abstract: Objective: To study the cytotoxic effect of dimethylhydrazine on PC12 and its mechanism of action. Methods: PC12 cells was incubated in Dimethyl hydrazine in the concentration of 600, 700, 800, 900, and 1 000 μmol·L-1 for 48 h. Then detected the inhibitory effect of Dimethyl hydrazine on PC12 cells proliferation in vitro by CCK-8 colorimetric assay. Comet assay showed the DNA damage in PC12 cells. The cell apoptosis was studied by flow cytometry. Results: Dimethylhydrazine has obvious toxic effect on PC12 cells after 48h, and the cell growth is obviously inhibited. Single-cell gel electrophoresis experiments showed significant DNA damage. All above had a significant difference compared with non-treat group(P<0.05). Conclusion: Dimethylhydrazine can significantly inhibit the proliferation of PC12 cells, damage cell DNA, and is dose-dependent. Its mechanism of action may be related to intracellular peroxidation.
