榛子花的质量标准研究

Research on Quality Standards of Corylus heterophylla Fisch

摘要: 目的:建立榛子花的薄层鉴别与榛子花中槲皮素、山柰素含量测定的高效液相色谱方法。方法:采用薄层色谱用硅胶G板对榛子花中槲皮素进行定性鉴别;采用色谱条件:色谱柱为 Sepax BR-C18(4.6 mm×250 mm,5 μm);流动相为甲醇-0.4%磷酸(48∶52);检测波长为360 nm;柱温:30 ℃;流速为1.0 mL·min-1。结果:采用的薄层色谱鉴别方法重现性好,专属性强。含量测定槲皮素、山柰素的线性良好(r>0.999 5);回收率分别为98.31% (RSD 1.3%)和96.80%(RSD 0.96%)。结论:建立质量标准方法简单、重现性好,为榛子花的质量控制提供测定依据。

Abstract: Objective: To establish a thin layer chromatography identification method for hazelnut flowers and to establish a HPLC method for the determination of quercetin and kaempferol in Corylus heterophylla Fisch. Methods: The quercetin in Corylus heterophylla Fisch was qualitatively identified on silica gel G plate of thin-layer chromatography. Chromatographic conditions were used: Inertsil ODS-SP C18 (4.6 mm×250 mm, 5 μm); mobile phase was methanol -0.4% phosphoric acid (48∶52) The detection wavelength was 360 nm; the column temperature was 30 ℃; and the flow rate was 1.0 mL·min-1. Results: The TLC identification method has good reproducibility and strong specificity. The linearity of quercetin and kaempferol in the content determination was good (r>0.999 5). The recoveries were 98.31% (RSD 1.3%) and 96.80% (RSD 0.96%), respectively. Conclusion: The establishment method is simple, prepared and reproducible, and can provide a reference for the application of the medicine.
