两种植物来源壮药鹰不扑小鼠急性毒性试验的比较研究

Composition and Toxicity Studies on Traditional Zhuang Medicine “Yingbupu”

摘要: 目的:比较两种植物来源壮药鹰不扑水提物、醇提物对小鼠急性毒性的影响,为民族医临床安全合理用药提供依据。方法:分别制备鹰不扑两种植物来源药材水提物、醇提物,按照经典小鼠急性毒性试验方法进行比较研究,测定小鼠口服半数致死量(LD50)、最大给药量,连续观察14 d,观察其急性毒性症状表现,记录累计死亡数。结果:两种植物来源鹰不扑对小鼠急性毒性强度为:虎刺楤木>黄毛楤木;不同提取物对小鼠急性毒性强度为:醇提物>水提物。虎刺楤木水提物、醇提物LD50值分别为99.0和57.4 g·kg-1,95%的可信限为66.2~148.0 g·kg-1、49.6~66.4 g·kg-1,分别相当于临床70 kg人用量的254.8倍和192.1倍。黄毛楤木水提物和醇提物无法推出LD50,最大给药量试验结果经计算均为100 g·kg-1。主要毒性症状是腹泻、怠动、毛色不华。结论:鹰不扑对小鼠具有一定的急性毒性,且其不同植物来源药材毒性差别较大。来源于虎刺楤木的药材急性毒性高于来源于黄毛楤木的药材,提示在临床使用中应根据药材来源确定使用剂量。

Abstract: Objective: To compare the acute toxicity of different components from two sources of traditional Zhuang medicine “Yingbupu” and to provide experimental data for safety use in clinical. Methods: Prepare the water extract and alcohol extract of the two plants of Yingbupu, and compare the acute toxicity on mice in accordance with classical acute toxicity test method. Results: The acute toxicity of Yingbupu from different sources was: Aralia armata (Wall.) Seem.> Aralia decaisneana Hance, and the acute toxicity of different components was: alcohol extraction > water extraction. The LD50 values of the water extraction in Aralia armata (Wall.) Seem. is 99.0 g·kg-1 and 57.4 g·kg-1, and the 95% confidence interval is 66.2-148.0 g·kg-1, which is equivalence to clinical 254.8 times of 70 kg people′s daily dried medicinal herb dose. The LD50 values of the alcohol extraction in Aralia armata (Wall.) Seem. is 57.4 g·kg-1, and the 95% confidence interval is 49.6-66.4 g·kg-1, which is equivalence to clinical 192.1 times of 70 kg people′s daily dried medicinal herb dose. The LD50 values of water extraction and alcohol extraction in Aralia decaisneana Hance can not be speculate, so the Maximum dose are calculated were 100 g·kg-1. Diarrhea, lazy, and uncoloured hair were the main acute toxic symptoms. Conclusion: The traditional Zhuang medicine “Yingbupu” have a certain acute toxicity, and the toxicity from different plants is quite different. The acute toxicity from Aralia armata (Wall.) Seem.was stronger than that from Aralia decaisneana. Hance, Prompt in clinical use should determine the dosage according to the plant of medicinal materials.
