羧基麦芽糖铁制备与质量特性表征方法研究

Preparation and Quality Characterization Methods of Ferric Carboxymaltose

摘要: 目的:研究羧基麦芽糖铁注射剂仿制药的质量控制方法,确保仿制药与原研药质量与药效的一致性。方法:采用改进的铁-碳水化合物制备方法制备羧基麦芽糖铁样品;采用动态光散射法测定羧基麦芽糖铁分子粒径,并辅助电镜法观察纳米颗粒;色谱法测定羧基麦芽糖铁的相对分子量,及糖基配体含量,原子吸收法测定铁含量;X射线粉末衍射法对铁核粒径进行检测,并均与原研厂羧基麦芽糖铁产品进行对比。结果:自制羧基麦芽糖铁样品和原研厂样品粒径、分子量、铁核粒径、糖基配体含量、铁含量结果接近,说明改进方法制备的羧基麦芽糖铁与原研药性质接近,同时,动态光散射法并辅助电镜法、色谱法、原子吸收法、X射线粉末衍射法可用于羧基麦芽糖铁的质量评价。结论:研究羧基麦芽糖铁质量评价的有效方法,能提高对羧基麦芽糖铁仿制药质量的控制,为该产品的质量和药效的一致性研究提供技术参考。

Abstract: Objective:To study the quality control method of ferric carboxymaltose injection and to evaluate the consistency of generic drugs. Methods:Ferric carboxymaltose samples were prepared by the optimized ferric-carbohydrate preparation method. The particle size of ferric carboxymaltose was determined by dynamic light scattering method and the nanoparticles were observed by electron microscopy. The relative molecular weight of ferric carboxymaltose was determined by chromatography, and the content of glycosylated ligand was determined by atomic absorption method. The particle size of ferric core was detected by X-ray powder diffraction, those compared with the ferric carboxymaltose original drug. Results: The particle size, molecular weight, iron core particle size, sugar ligand content and iron content of the self-made ferric carboxymaltose sample were similar to those of the original sample, indicating that the properties of ferric carboxymaltose prepared by the improved method were similar to those of the original drug. Meanwhile, dynamic light scattering, electron microscopy, chromatography, atomic absorption spectrometry and X-ray powder diffraction can be used to evaluate the quality of ferric carboxymaltose. Conclusion: Effective method to control ferric carboxymaltose quality evaluation, is effective to improve the quality control of generic drugs of ferric carboxymaltose, to ensure the quality of generic drugs and efficacy of consistency with the original drug, and provide technical reference for the consistency research of quality and efficacy of the generic drugs.
