复方大青叶合剂指纹图谱的建立与分析

Establishment and Analysis of Fingerprint Spectra of Compound Daqingye Mixture

摘要: 目的:建立复方大青叶合剂的指纹图谱,为其质量控制提供依据。方法:采用HPLC法测定,色谱柱为岛津Shim-pack GIST C18(250 mm×4.6 mm,5 μm),流动相为甲醇-0.02%磷酸,梯度洗脱,流速为1.0 mL·min-1,检测波长为260 nm和326 nm。结果:共标定出20个共有峰,并通过对照品比对指认其中8个成分,分别为腺苷、没食子酸、绿原酸、紫花前胡苷、芦荟大黄素、大黄酸、大黄素、大黄酚。不同厂家之间以及相同厂家不同批次制剂相似度均存在差异。结论:该研究建立的HPLC指纹图谱具有良好的稳定性和重复性,为复方大青叶合剂的质量控制提供了参考。

Abstract: Objective: Establish a fingerprint of Compound Daqingye Mixture to provide a basis for its quality control. Methods: HPLC method was used for determination, with Shim-pack GIST C18 as the chromatographic column(250 mm×4.6 mm, 5 μm). The mobile phase is methanol-0.02% phosphoric acid, gradient elution, flow rate is 1.0 mL·min-1, and detection wavelengths are 260 nm and 326 nm. Results: A total of 20 common peaks were identified, and 8 components were identified through comparison with reference materials, including adenosine, gallic acid, chlorogenic acid, purple flower front saponin, aloe emodin, rhein, emodin, and chrysophanol.There are differences in the similarity between different manufacturers and different batches of formulations from the same manufacturer. Conclusion: The HPLC fingerprint established in this study has good stability and repeatability, providing a reference for the quality control of Compound Daqingye Mixture.
