HPLC测定大鼠肠道菌群中辛伐他汀及其代谢物的方法学研究

Determination of Simvastatin and Its Metabolites in Intestinal Microbiota of Rats by HPLC

摘要: 目的:建立HPLC测定大鼠肠道菌群中辛伐他汀及辛伐他汀酸浓度的方法。方法:取含辛伐他汀或辛伐他汀酸的大鼠肠道菌群孵育液加入含有内标物(异替格瑞洛,浓度为:1×103 ng·mL-1)的蛋白沉淀剂沉淀,离心后取上清液进行HPLC分析。色谱条件:Amethyst C18-H色谱柱(4.6 mm×250 mm,5 μm);流动相甲醇(A)-0.1%磷酸溶液(B)(80∶20)。检测波长237 nm,流速 0.8 mL·min-1,柱温 30 ℃,进样量50 μL。结果:采用甲醇溶液作为蛋白沉淀剂,含药菌液与甲醇溶液比例为1∶2时,在4 ℃下离心力为15 250 g离心,对辛伐他汀及辛伐他汀酸提取率最高。结论:该方法专属性强、提取率高、精密度好、操作简便,可应用于辛伐他汀相关的药动学研究。

Abstract: Objective:To establish a HPLC method for the determination of simvastatin and simvastatin acid in intestinal flora of rats. Methods: The intestinal microbiota incubation solution containing simvastatin or simvastatin acid was precipitated by adding protein precipitator containing internal standard(isoticagrelor, concentration: 1×103 ng·mL-1). After centrifugation, the supernatant was taken for HPLC analysis. Chromatographic conditions: Amethyst C18-H column(4.6 mm×250 mm, 5 μm); Mobile phase methanol (A)-0.1% phosphate solution(B)(80∶20). The detection wavelength was 237 nm, the flow rate was 0.8 mL·min-1, the column temperature was 30 ℃, and the injection volume was 50 μL. Results: The extraction rate of simvastatin and simvastatin acid was the highest when methanol solution was used as protein precipitator, the ratio of drug-containing bacteria solution to methanol solution was 1∶2, and the centrifugal force was 15 250 g at 4 ℃. Conclusion: This method has strong specificity, high extraction yield, good precision and simple operation, and can be applied to the pharmacokinetic study of simvastatin.
