Research on the Determination of Aflatoxin B1 Content in Sterculiae Lychnophorae Semen by Standard Addition Method

摘要: 目的:采用标准加入法对中药胖大海中的黄曲霉毒素B1进行含量测定,并分析该方法的优点和局限性。方法:使用高效液相-柱后光化学衍生法为检测方法,采用标准加入法分析胖大海中的黄曲霉毒素B1的含量,为验证该方法的准确性与可靠性,使用超高效液相-串联质谱法(UPLC-MS/MS)对同批样品进行平行测定,并对两种方法的结果进行比对分析。结果:标准加入法能够减少样品前处理过程中带来的损失和干扰,有效抵消基质效应,方法结果准确、可靠,但该方法样品消耗大、对实验操作和线性范围要求较高,在批量分析、样品量有限等条件下存在一定的不足。结论:使用标准加入法可有效克服基质干扰的影响,该法可作为标准曲线法的补充,有效提高胖大海中黄曲霉毒素B1含量测定的准确性,但也存在操作和适用性上的局限。

Abstract: Objective: The content of aflatoxin B1 in Sterculiae Lychnophorae Semen was determined by the standard addition method, and the advantages and limitations of this method were analyzed. Methods: Using high-performance liquid chromatography with post-column photochemical derivatization (HPLC-FLD) as the detection method, the standard addition method was analyzed the content of aflatoxin B1 in Sterculiae Lychnophorae Semen. To verify the accuracy and reliability of this method, the same batch of samples were simultaneously determined by ultra-performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS), and the results of the two methods were compared and analyzed. Results: The standard addition method can reduce the loss and interference caused by the sample pretreatment process and effectively offset the matrix effect. The method is accurate and reliable. However, the method also has some shortcomings, such as large sample consumption, high requirements for experimental operation and linear range, making it less suitable for batch analysis, and scenarios with limited sample availability. Conclusion: The standard addition method can effectively overcome the influence of matrix effect. This method can be used as a supplement to the external standard method to effectively improve the accuracy of the determination of aflatoxin B1 content in Sterculiae Lychnophorae Semen, though it still presents certain limitations in terms of operational complexity and applicability.
