注射用两性霉素B脂质体细菌内毒素检查方法的评价

摘要: 目的:建立准确、可靠的注射用两性霉素B脂质体细菌内毒素检查方法,以解决现行质量标准中出现假阳性结果的情况。方法:采用两个厂家生产的动态显色法鲎试剂进行检测,将结果进行比对;同时尝试使用动态浊度法和凝胶法进行试验。结果:采用动态显色法对注射用两性霉素B脂质体进行内毒素含量检测,两种鲎试剂检查结果存在明显差异,易出现假阳性的情况。采用凝胶法或动态浊度法进行测定,实验结果均符合规定要求。结论:注射用两性霉素B脂质体为有色制剂,其颜色与动态显色法反应终点颜色相近,存在颜色干扰风险。因此,在该品种细菌内毒素检查方法的选择中,更推荐选择动态浊度法或凝胶法。若使用动态显色法检测,不同生产企业生产的鲎试剂的检查结果可能存在差异,且假阳性风险较高。为避免此种情况,建议选择动态浊度法或凝胶法。采用动态浊度法和凝胶法对注射用两性霉素B脂质体细菌内毒素进行检查,结果较动态显色法更为可靠、准确。

Abstract: Objective: To establish an accurate and reliable method for bacterial endotoxin testing of amphotericin B liposomes for injection, in order to solve the problem of finding an accurate and effective method for bacterial endotoxin testing of amphotericin B liposomes for injection in current quality standards, and to address the issue of false positive results in existing quality standards. Methods: Two manufacturers′ dynamic colorimetric horseshoe crab reagents were used for inspection and testing, and the results were compared. At the same time, the dynamic turbidity method and gel method are tried to be used for the test. Results: The dynamic colorimetric method was used to detect the endotoxin content in amphotericin B liposomes for injection. There was a significant difference in the results of the two horseshoe crab reagents, and false positives were prone to occur. The gel method or dynamic turbidimetric method are used for determination, and the experimental results meet the specified requirements. Conclusion: Amphotericin B liposomes for injection are colored preparations, and their color is similar to the endpoint color of the dynamic colorimetric method, which poses a risk of color interference. Therefore, in order to avoid color interference, the dynamic turbidimetric method or gel method is usually recommended for endotoxin test. If dynamic colorimetric method is used for detection, there may be differences in the test results of horseshoe crab reagents produced by different manufacturing enterprises, and the risk of false positives is high. False positive results are also prone to occur. In order to avoid this situation, it is recommended to select dynamic turbidity method or gel method. The test results showed that the dynamic turbidimetric method and gel method were more reliable and accurate than the dynamic color display method in the detection of bacterial endotoxin in amphotericin B liposomes for injection. The dynamic color rendering method is more reliable and provides accurate results.
